INTERVENTION: Intervention1: Green tea leaves: Standardized laboratory‐prepared Green tea leaf extract will be used as the root canal irrigant during biomechanical preparation. A total volume of 5 mL will be used for 5 minutes. Microbial samples will be collected before biomechanical preparation (S1) and immediately after biomechanical preparation (S2) using sterile paper points for RT‐PCR analysis. Intervention2: Neem: Standardized laboratory‐prepared neem leaf extract will be used as the root canal irrigant during biomechanical preparation. A total volume of 5 mL will be used for 5 minutes. Microbial samples will be collected before biomechanical preparation (S1) and immediately after biomechanical preparation (S2) using sterile paper points for RT‐PCR analysis. Intervention3: Coriander oil: Standardized laboratory‐prepared Coriander oil extract will be used as the root canal irrigant during biomechanical preparation. A total volume of 5 mL will be used for 5 minutes. Microbial samples will be collected before biomechanical preparation (S1) and immediately after biomechanical preparation (S2) using sterile paper points for RT‐PCR analysis. Control Intervention1: 3 percent Sodium hypochlorite: Commercially available 3 percent sodium hypochlorite will be used as the root canal irrigant during biomechanical preparation. A total volume of 5 mL will be used for 5 minutes. Microbial samples will be collected before biomechanical preparation (S1) and immediately after biomechanical preparation (S2) using sterile paper points for RT‐PCR analysis. CONDITION: Health Condition 1: K041‐ Necrosis of pulp Health Condition 2: K044‐ Acute apical periodontitis of pulpal origin PRIMARY OUTCOME: Reduction in Enterococcus faecalis bacterial load following biomechanical preparation using the allocated root canal irrigant, quantified by RT PCR.Timepoint: At Baseline and immediately after completion of biomechanical preparation and irrigation during the first same treatment visit. SECONDARY OUTCOME: Reduction in postoperative painTimepoint: 24hrs 48hrs 72 hrs INCLUSION CRITERIA: Patients with age group of 18 to 40 years of either gender presenting with necrotic pulp with symptomatic apical periodontitis of maxillary central incisors with periapical inde Xscore of 2 to 4 Patient with preoperative pain scores as severe score of 7 to 10 on a visual analog scale. Patients with no systemic disease. Periodontally healthy individuals. Patients with good oral hygiene. Patients willing for voluntary participation & have signed informed Consent.
