The invention discloses a siRNA of interfering with gene expression of a COVID-19. The siRNA is one of the following double-stranded RNA sequences: a double-stranded RNA sequence with a positive-sensestrand SEQ ID NO:5 in a sequence table and an antisense strand SEQ ID NO:6 in the sequence table, a double-stranded RNA sequence with a positive-sense strand SEQ ID NO:7 in the sequence table and anantisense strand SEQ ID NO:8 in the sequence table, and double-stranded RNA sequences which have the similarity of 95% or above to the defined double-stranded RNA sequences and have a function of silencing an RDRP gene or an S gene. The siRNA disclosed by the invention is designed for mRNA sequences of an S protein and an RDRP enzyme on the COVID-19; expression of a key protein of a virus can be effectively inhibited; and important reference significance is provided for development of an anti-COVID-19 drug.
